  1. One thousand bases of quail mitochondrial DNA were downloaded to Geneious from the GenBank database. PCR primers were designed to amplify a conserved region from this sequence. Each primer was 20 bases long, and the 5` end of the forward primer (F) and reverse primer (R) starts from base position 33 and 865, respectively. A PCR master mixture for 23 reactions to analyse different bird samples was prepared.

One difficulty in extracting sequencing reads that correspond to mitochondrial DNA from mixed fragments of nuclear and mitochondrial DNA is the nuclear genome contains segments homologous to regions of the mitochondrial genome called "numts." Mammalian genomes contain 50-450 kb of numts (the human genome contains 1005 numts, averaging 446 bp). Estimate the fraction of reads from fragments of mammoth DNA that are likely to be numts. The mammoth genome is approximately 4.7x10^9 bases in length.

You are setting up a PCR reaction using DNA from a bacteria with a genome size of 17 megabase-pairs (mb). How many nanograms of DNA will you need to add to the reaction to ensure there are at least 100 copies of the single gene you are trying to amplify? (assume the average molecular weight of a nucleotide pair is 650). (2 pt)

Thailand is one of the most popular locations for Chinese tourists.  There were about 70 thousand Chinese tourists visited Thailand every year.  The competition among Thai tourist agents is very fierce. Some agents continue to cut their prices until the price is zero.  This phenomenon is called “zero-dollar tour package”. At the same time there often news reports that many tourist agents often take the tourists to fake jewelry shops.  Use the theory about market failure to explain this zero-dollar...

One thousand cars were stopped at random for a roadside test of tyres and lights: 115 had unroadworthy tyres; and 46 had a lighting fault. These figures include 22 cars with both defects. If drivers had been fined $100 if their car had one defect, and $500 if their car had both defects, how much revenue would have been raised per car?

Researchers examined the relationship between cola consumption and bone loss after fifty. One thousand women were randomly sampled, everyone's cola consumption was recorded over a two month period of time, and bone density measures were also recorded. Researchers found that the more cola consumed, the lower the level of bone density. They concluded that cola consumption leads to loss of bone density. What type of study is this research and why? Identify the variables. Evaluation the internal validity. Researchers examined...
