Compare and contrast GPCR signaling and enzyme-coupled signaling. Focus on the general mechanism of these signaling pathways.

Solutions

Expert Solution

Ans) 1) The enzyme linked coupled receptor has intrinsic tyrosine kinase which will autophosphorylate the tyrosine residues which then serves as a binding site for Grb2 protein wheras GPCR do not have such intrinsic tyrosine kinase associated.

2) In receptor tyrosine pathway, PKB (protein kinase B) is activated by PIP3 kinase. PKB can also be activated by IRS-1.Then the activated PKB will activate AKT kinase which inturn activate BAD protein which causes apoptosis.Wheras in GPCR pathway, PKA (protein kinase A) is activated by cAMP which activate CREB (cAMP response element binding protein) which enters nucleus to turn ON the cell cycle. PKC (Protein kinase C) can also get activated by the DAG (diacylglycerol).

Similarities between both of these pathways is that after the primary ligand binding, the conformational change occurs in the receptor. Another similarity is that both of these pathways can turn ON the cell cycle.
