coupled-enzyme assay
The determination of a substrate or enzyme activity by coupling of
one enzymic reaction with another, more easily detectable,
reaction.
The product of the first reaction is the substrate for the second.
These assays are often designed to generate or consume a reduced nicotinamide-containing nucleotide, which is measurable by its ultraviolet absorption.
Even when the enzyme reaction does not result in a change in the absorbance of light, it can still be possible to use a spectrophotometric assay for the enzyme by using a coupled assay.
Here, the product of one reaction is used as the substrate of another, easily detectable reaction.
Bellow is the example of coupled assay.
