8. Why does the extract have to be concentrated prior to analysis for low concentration samples?

9. Why is sample clean-up necessary?

Solutions

Expert Solution

8.The development of new LC-MS/MS methods for small molecules in biological fluids is becoming increasingly more challenging, because of the need to continuously achieve higher sensitivity and better assay robustness in complex biofluids such as serum, plasma, urine, oral fluid or cerebrospinal fluid (CSF). In addition, because of the very low concentration levels of( pharmaceutical) targets, samples often need to be pre-concentrated before analysis. Unfortunately, this does not only increase the concentration of the desired compound in the sample extract but also often raises the levels of interfering components. As a result, very specific and effective sample clean-up procedures are required for sensitive and selective LC-MS/MS assays.

9.The primary goal of sample preparation is to isolate one or several target analytes from the other components of the sample mixture (matrix). Depending on their nature and concentration levels, co-components of the sample matrix can influence the quantitation of target analyte(s) during subsequent liquid chromatography-mass spectrometry (LC-MS) or tandem mass spectrometry (LC-MS/MS) experiments if not removed prior to analysis.

Suppose you have a concentrated solution (S1) with a concentration equal to 1 M. You use their solution to make more dilute solutions S2-S6. Complete the following table, which describes the solution process. NOTE: Not all of the solutions are diluted S1; some are made from solutions that are derived from S1. The equation C1V1 = C2V2 should be used as a starting point for calculation. You can assume all volumes are additive. S2 is shown as an example. Solution...

mass spectral measurements were made on 10 replicate low concentration Ni-EDTA samples: 146.9 , 111.5 , 125.7 , 140.7 , 109.7 , 179.1 , 130.7 , 135.3 , 124.7 , and 142.3 counts. Ten measurements of a blank had a mean measurement of 38.3 counts. A sample containing 2.00 μM Ni-EDTA gave a measurement of 1597 counts. Estimate the signal and detection limit for Ni-EDTA.
