Immunoassay precipitation reactions are done by either detecting the antigen or by detecting the antibody. The technique by which unknown antigen is detected is the precipitin ring assay or the sandwich ELISA. In both the assays known monoclonal antibodies against the antigen are prepared and added in the solution. In the precipitin ring assay a precipitin ring is formed when the equivalence zone is formed. In ELISA the antigen is detected by adding another flourescent secondary antibody which changes the color of the solution when a substrate is added.
The technique by which the antibodies are detected is the Direct ELISA in which the antigen is coated in the well and the sample is added into the well. If the antibodies that bind to the antigen are present in the samples, then they are detected and hence the antibodies are detected. Hope this is what you were asking in the question.
