what is the primary metabolite guaifenesin is metabolized by O-demethylase?

Solutions

Expert Solution

Chemical name - 3-(2-methoxyphenoxy)-1,2-propanediol

molecular formula -C10H14O4 with a molecular weight of 198.21

Appearance- white or slightly gray crystalline substance

Solubility - freely soluble in ethanol.

Absorption- readily absorbed in GI tract

Function - an expectorant by helping to loosen mucus

Metabolisation- Guaifenesin experiences rapid hydrolysis and also experiences both oxidation and demethylation . It is quickly metabolized in liver by way of oxidation to β-(2-methoxyphenoxy)-lactic acid and demethylated by O-demethylase in liver microsomes to the point where it is excreted as this metabolite in the urine within few hours. O-demethylase is the primary enzyme for the metabolism of guaifenesin.

The primary metabolites of the substance are β-(2-methoxyphenoxy)-lactic acid and the demethylated hydroxyguaifenesin, both of which are themselves inactive compounds
