You have made 1L of a 0.1M HEPES buffer, and now you need to adjust the pH to 7.7. Calculate how much of a 0.5 M NaOH solution you would need to add. I was told to use Henderson-Hasslebach, I'm not sure how to do this one

Solutions

Expert Solution

HEPES pKa = 7.5

pH = 7.7

moles of HEPES = 0.1 x 1 = 0.1

moles of NaOH = 0.5 x V

HEPES + NaOH --------------------------> salt

0.1              0.5 V                                    0 -------------------> initial

0.1-0.5 V      0                                        0.5 V ------------------> after reaction

Henderson-Hasslebach

pH = pKa + log [salt / HEPES]

7.7 =7.5 + log (0.5 V / 0.1 -0.5 V)

1.585 =(0.5 V / 0.1 -0.5 V)

-0.792 V + 0.1585 = 0.5 V

1.292 V = 0.1585

V = 0.1226 L

volume of NaOH =122.6 mL needed

Prepare 100mL of a 0.1M potassium phosphate buffer of pH 6.5. You have K2HPO4 and KH2PO4 in solid/salt form available to mix with water to create this buffer. Final answer must include how many grams of each salt to mix with how many mL of water to prepare the buffer. I started this problem and got as far as using the Henderson-Hasselbalch equation and that the ratio of [HPO42-] to [H2PO4-] is 0.48 moles to 1 mole.

Assume you have successfully made a buffer solution, such that you have 0.250 M phosphate buffer at pH 7.2. You dissolve in this solution sufficient histidine (side chain pKa = 6.0), such that the final histidine concentration is 5 mM, all at 25°C. Therefore, the pH of the solution is controlled by the phosphate buffer. In this solution, what is the percent of histidine with a protonated side chain? What is the total charge on molecules in this fraction? Explain.
