Polymerase chain reaction technique use to copy of  DNA using small amount of sample, DNA sequences are amplified in a series or cycles of temperature changes. In other words It can be said  that it is in vitro replication process.

There are three steps of PCR:

A . Denaturation

In natural replication process it is necessary to separate double stranded to copy each strand same in PCR denaturation of DNA carried out by means of temperature. It separates complementary nucleotide.

DNA sample treated for 30 seconds at 90 degree. It helps to break hydrogen bond.

B. Annealing

Primer used for annealing purpose. A small known sequence of DNA direct for amplification process. The test tube set to 50 degrees Celsius for 30 seconds.

C. Extension

In this step the addition of nucleotide to complimentary is proceed. For polymerization reaction taq polymerase used. Taq polymerase isolated from Thermus aquaticus thermophilic bacteria & process carried out at 70 degree Celsius for 60 seconds.
