The 1st recombinant DNA molecule was made by Boyer & Cohen in 1973.

Any DNA molecule from any orgnaism or artificially made DNA can be inserted to make a recombinant DNA molecule, as DNA is afterall DNA.

Restriction enzymes and ligases help to cleave of the gene of interest and ligate with any DNA sequence.

Class II restriction enzymes are of value to genetic engineering and recognize a palindromic sequence for cleavage. They can create either sticky ends or blunt ends. Ligation of sticky ends is easier than that of blunt ends.

Following steps are involved in creating a recombinant DNA molecule and eventually a recombinant protein -

  • Identification of gene of interest.
  • Isolation of genes.
  • Insertion of the isolated gene into a vector.
  • Transformation into a host.
  • Identification of recombinant clone.
  • Growth of the recombinant clone.
  • Isolation and purification of recombinant protein.
