What are the pharmacodynamics of Cocaine Hyrdrochloride?
Pharmacokinetics:
Absorption:
Oral doses are absorbed in 30 mins while maximum concentrations is reached in 50 to 90 mins.
inhalation dose is absorbed in 3 mins while peak will come in 10 to 15 mins.
In alkaline medium it is well absorbed.
Onset of action in brain is 8 to 12 seconds.
Distribution:
It is distributed in all body tissues and it crosses blood brain barrier. Since it is soluble in lipids it
is high in CNS and adipose tissues.
Half life in oral route is 0.7 hrs.
in parenteral route is 0.8 to 09 hrs
in nasal route is 1.25 hrs
Metabolism:
Metabolism occurs in liver within 2 hrs rate depending on the plasma concentration
Elimination:
Most of the drug eliminated in 4 hrs. whereas metabolutes will be in plasma for 144 hours.
It is excreted in stools and saliva too
Pharmacodynamics:
Action is sympathetic one and it prevents the reuptake of the dopamine and noradrenaline leading to stimulation of neurotransmitters. It also prevents the serotonin release. When applied locally provides anesthetic action by blocking the neuronal transmitters.
