Q5. When do we use the ED50 and EC50, instead of the LD50 and LC50?
ED 50 : Median effective dose, it is the dose required to achieve the 50 percent of the desired response in the population.
EC 50 : half maximum effective concentration, it is the concentration of the drug at which 50 percent of its maximum response is observed.
LD 50 : Median lethal dose, the dose required to achieve the 50 percent mortality of toxicity.
LC 50 : Half lethal concentration, the concentration of drug at which 50 percent of mortality from toxicity is observed.
During pre clinical studies of a drug on animals to determine the toxic dose of the drug we use LD 50 and to determine maximum safety profile of a drug we use LC 50 in animal studies.(acute, sub acute and chronic toxicity studies).
During clinical trials to determine the therapeutic window of the drug ED 50 that is 50 percent of the drug dose is efficient to produce the desired therapeutic response in the population were as EC 50 effective concentration that is at 50 percent of drug dose we can achieve the maximum therapeutic effect (for e.g in case of patients who are fast metabolizer's). During phase 1 to define the dose in clinical trials and safety profile in phase 2 trials.
