List 4 safety features that you will need to follow when using the pipette (In microbiology).

Solutions

Expert Solution

Do not pipette by mouth (avoid mouth-pipetting). Use pipette bulbs or pipetting devices for the aspiration and dispensing of liquid cultures.

Label all pipettes clearly. All cultures and medium should be clearly labeled with names and date of use. Incase they are hazardous, proper warning and hazard information should also be labelled

Wear Personal protection equipments (labcoat, eye wear and gloves) - example disposable gloves to avoid hand contamination or avoid infection in wound or cut especially while working with potentially infectious microbes

Sterilised pipette should be used to transfer cultures, and avoid contamination

Hold pipette vertically when drawing liquid cultures. Also, pipette slowly and smoothly

Avoid touching tip of the pipette, to avoid contamination

Do not insert the pipette into the hole in the bulb, as the pipet might break. Use great care while inserting pipette in the bulb or use suction device

Report spills immediately

In a lab you are asked to pipette 130 microL using a pipette with a range of 20-200 microL. This pipette has recently been calibrated so is functioning correctly. You correctly move the dial of the pipette to 130 microL. On your first attempt to dispense 130 microL, you realise that the volume expelled was only 105 microL. On the second occasion it was 170 microL. Give explanations to what operator errors could have led to these outcomes?

Give the most accurate value for the number of viable cells in the original stock culture, in CFU / ml. Just enter a number,...
