A species of mountain dragon is being studied. Mountain dragons have blues scales and blue blood. The blue colour of their scales is caused by a pigment protein produced only in the scale cells. Dragons do not use hemoglobin to transport oxygen like we do, they use something called dragocyanin. Our blood appears a reddish colour due to the iron contained within the hemoglobin complexes. Dragon blood is blue because dragocyanin contains copper instead of iron. Dragocyanin is only produced within the blood cells of dragons. If you wanted to sequence the DNA of the blue pigment molecule that determines scale colour what tissue or cell type could you use (in other words, where could you collect the DNA from in order to sequence it)? Explain your choice.You measured the amount of DNA methylation on the blue pigment gene promoter and the dragocyanin gene promoter, in scale cells, blood cells, and muscle cells. What do you expect from your results? For your answer you can just use "low" and "high" to describe the amount of DNA methylation. For example (using different genes): In scales the promoter of gene X will have high level of methylation, in blood a low level, and in muscle cells a low level.
In order to sequence the DNA of the blue pigment molecule we will take the DNA of the scale cells as it is in these cells the DNA is completely functional and acted upon by transcriptional machinery to transcribe the DNA and then it gets translated into the blue pigment that renders the scales their blue colour.
DNA methylation is a repressive mark on the chromatin and denotes silenced genes to prevent their transcription. This chromatin modification is highly tissue specific and varies from low to high in different tissues.. The tissue in which the DNA methylation over a particular gene promoter will be low will easily transcribe and on which it will be high will not transcribe.
For the blue pigment that transcribes only from the scale cells:
