In UV-VIS, list and describe 5 things that you might change to reliably to be able to detect lower concentration levels of sample.
1) It is essential to be absorbance lies in between 0.1 to 1. If the absorbance is found to be greater than 1, then it is a high concentration solutions and then it will not follow the Lambert beer law. We need to dilute the sample until the absorbance is comes upto 1.
We need to prepare the various known concentrations solutions and check the absorbance through uv . The graph is plotted between absorbance and concentration and then find out the slope by fitting straight line curve. This slope is further use to find the concentration of solutions at different time. Obviously concentration is decreases with time and hence absorbance.
There is a need to absorbance lies in between 0.1 to 1 otherwise curve is not proper and spectrum overlap.
Cautiously prepare the solutions
Resolution should maintain properly.
After preparation instant checking in UV is recommended otherwise other kind of reaction is start.
Calibration curve is proper.
