4. Which of the following statement about chaperones is TRUE? (0.2 pt) Please select the single best answer and explain why other answers are wrong (0.8 pt).

  1. HSP100 unfolds misfolded proteins and degrade them.
  2. HSP100 activity requires the ubiquitination of the client proteins.
  3. HSP100, but not HSP70, requires ATP to execute their chaperones functions.
  4. HSP70 binds to client proteins through electrostatic interactions.
  5. HSP70 can deliver client proteins to other chaperones.

Solutions

Expert Solution

4.E   HSP70 can handover client proteins to other (co)chaperone machines for further folding or degradation and this will be further referred to as an external HSP70 network. Hsp90 binds client proteins through an extended interface that enables a large number of low-affinity contacts. The heat shock protein 70 (Hsp70) chaperones, vital to the proper folding of proteins inside cells, consume ATP and require cochaperones in assisting protein folding. Hsp70 play key roles in the triage of damaged and aberrant proteins for degradation via the ubiquitin-proteasome pathway. In the mid 1990s, it was discovered that Hsp70 is required for ubiquitin-dependent degradation of short lived and abnormal proteins,Hsp100 chaperones support heat-shock survival of bacteria, yeast, and plants. Hsp100 reactivate aggregated proteins in cooperation with Hsp70/40. Hsp100 chaperones (ClpB/Hsp104) belong to the AAA+ family of ATPases. Hsp100 use energy from ATP to extract single polypeptides from aggregated particles.
