The cSMAC or central supramolecular activation complex contains receptor proteins TCR, CD4, CD8, CD28, and CD2. This structure is required for activation of T cells.
The pSMAC or peripheral supramolecular activation complex is mainly composed of integrin molecule lymphocyte function-associated antigen (LFA) -1. The pSMAC makes the initial interaction with the target cell as the integrin protein binds to the target cell’s ICAM-1 or ICAM-2.
The dSMAC or distal supramolecular activation complex contains the actin structures such as lamellae and lamellipodia, and CD43 and CD45 molecules. This part is essential for scanning the surface of the target cell, which thereby assists the integrin of pSMAC.
