Our set of coloured calibration standards can now be transferred to 1 cm cells (shown below) to perform UVvis absorption spectrophotometry in a double beam instrument at the λmax value. What reagent do you use for the reference cell in the double beam spectrometer?
Reagent should be one which follows the following criteria:
1. As mentioned in the question, the caibration standards are colored which means the reference cell should also have a colored solution which is of a concentration higher than the standards' concentration.
2. Another important point while taking the reagent is that the absorbance for the reference solution is set to 1 or equivalent to 100 % or in other words, transmittance set to zero.
3. The reagent taken for the UV-visible spectrometry experiement should be chemically inert, in a way that the chemical form (or formula) does not change during the course of the analysis.
