Write a case you can use PCR to solve the problem and also write a case you cannot use PCR to solve to problem

Solutions

Expert Solution

PCR is polymerase chain reaction used amplify gene of interest. In order to solve crime cases, an allele is chosen which is amplified to look for differences between the samples of the crime scene and that of the suspects. This is usually done when the allele varies in size because of repeats in different individuals. This is known as Short Tandem Repeat (STR) which is used to compare allele repeats at specific loci in DNA between two or more samples. The PCR product is run on gel to compare the size of bands. More the repeats, more is the size of DNA and less it travels from the well of the gel. This is an example where PCR can be used to solve the problem.

AN example where PCR cannot be used to solve a problem is in case of single-nucleotide polymorphism (SNP) which is a DNA sequence variation that occurs at a single nucleotide adenine (A), thymine (T), cytosine (C), or guanine (G). This cannot be distinguished on a gel after running the PCR product because SNP does not lead to different sized band.

(Use the string class to solve the problem) Write a program (in c++) that can be used to train the user to use less sexist language by suggesting alternative versions of sentences given by the user. The program will ask for a sentence, read the sentence into a string variable, and replace all occurrences of masculine pronouns with gender-neutral pronouns. For example, it will replace “he” with “she or he”, and “him” with “her or him”. Be sure to preserve...

In this problem, you can use the Matlab program posted on course website and Canvas (also given in the lecture) that computes the interpolation polynomial. We want to see how well a given function can be approximated by the interpolation polynomials. Let f be a function. We divide the the interval [−0.6,0.6] into subintervals of the same length h = 0.02. The gridpoints are −0.6 = x1 < x2 < ... < x61 = 0.6. Take N = 61 points...

Describe how you can use PCR to obtain a gene fragment X that can be cloned into a plasmid vector for expression of the recombinant protein. Include in your description how the PCR reaction will be performed, how the amplicon will be analyzed, the use of restriction enzymes, how cloning will be achieved, how you will know that you have a recombinant containing the insert gene fragment X and how you will detect the recombinant protein.
