Why are these materials typically used ?? Please answer for both materials please.
1.Reagent kit for Probe Labeling
2.Oligonucleotides for PCR amplification
1. Reagent kit for prob labeling contains reagents for labeling the probes. A probe is a single stranded piece of DNA labelled either with radioisotopes or with nonradioactive material. The nucleotide sequence of probe is complimentary to the target DNA. A probe can identify a specific DNA fragment within a DNA library through base pairing. For example a biotin labeling reagent kit contains biotin ragent, tris buffer, sec-butanol, precipitant and biotinylated DNA standard. A reagent kit is used to label properly a DNA probe without altering its sequence.
2. Oligonucleotides are used as primers in PCR amplification of target DNA. Such primers are generally between 18 and 30 bases long so that the primer sequences can amplify a unique sequence in the target genome. The primer binds to the specific DNA sequence and DNA polymerase is used to extend the oligonucleotide and replicate the complementary strand.
