The following are the steps are to be followed for the dissection of rat brain

  • Sacrifice the rat as per the ethical rules. and remove the rat brain .
  • Wash the rat brain with cold water and add DEPC .
  • DEPC is used to remove any blood stains on the brain.
  • On a metal plate , cut the brain into half . Separate the brain into left and right hemispheres.
  • Remove the olfactory lobe and place it in the liquid nitrogen .
  • Store the specimen at -80 degree celcius.
  • Hippocampus is present in the mid brain. Now for collecting the hippocampus , the ventral side of the brain is placed on the top .
  • The hippocampus is removed from the mid brain and place store at -80 degrees in liquid nitrogen .
  • In the case of any neuro degenerative disease, the ER protein synthesises defective protein.
  • To measure the amount of ER protein immunoenzymatic assay is followed.
  • The tissue obtained is treated with high levels with oestrogen .
  • If the protein is normal , then there will be more amounts of oestrogen binding sites. If it is abnormal the binding sites will be less .
  • A monoclonal antibiody is allowed to bind to the protein that contains oestrogen and measured for the amount of protein present in the tissue.
  • To construct DNA libraries , the tissue is treated with poly A RNA.
  • Plus/minus screening of cDNA libraries are followed.
  • Inorder to identify a specific expression in case of normal and abnormal tissue , probes or p[rimers that are complementary are used.
  • Antibody complementary to that particular probe is used to isolate the DNA or protein.
  • For isolating DNAS , this method is called nucleic acid hybridisation .
  • Thus, proteins or DNAs can be isolated specifically in Rat brain.
