The following data was generated for an isolated mammalian dihydrofolate reductase.

[7,8-dihydrofolate] (mM)          V (mol/s)                V (mol/s) with methotrexate

                1.1                                  333                                      169

                2.0                                  444                                      265

                5.0                                  667                                      465

A.        Graph the data with and without methotrexate on a Lineweaver-Burk plot. Clearly label which line is which.

B.        Calculate Km for the enzyme in the presence and absence of methotrexate. Is this a competitive or non-competitive inhibitor?

C.        Considering that deoxynucleotide concentrations are limiting in the cell, what effect will addition of methotrexate have on DNA replication? Explain your reasoning.

Solutions

Expert Solution

Theory of Lineweaver-Burk plot.

a.

b. by the theory if you extrapolate the lines you can find the Km. the results are:

With meth: -1/km= -0.6 Km=1.666

Without meth: -1/km = -0.25 Km=4

the case is the same as the first one. Which is Competitive inhibition.

For c. this is more a theory question, I dont have the enogh data.
