A sample mixture of ethyl benzoate (bp 212 C) and dodecane (bp 216.2 C) is injected on two GC columns. Column A has DC710 silicone oil as the stationary phase, and column B uses polyethylene glycol as the stationary phase. Which substance would be certain to elute first from column A and would the same material be expected to elute first from column B? Which column, A or B, would be expected to give the better separation of these two substances?
In GC, the stationary liquid phase controls the separation of compounds based on two criteria. If there is no interaction between the sample and stationary phase, the boiling point of the material determines the order of elution and the component with the lowest boiling point will elute first from the column. It the functional group of the sample interacts with the stationary phase, then the elution depends on the binding properties. the sample with less affinity towards stationary phase elutes first.
In the case of Column A DC710 Silicon Oil as stationary phase, the separation is based on the boiling point. therefore ethyl benzoate (bp 212 0C) eluted first followed by (216.2 0C)
In the case of column B polyethylene glycol as the stationary phase, the elution depends on the binding properties. Thus dodecane is expected to elute first from column B due to the greater affinity of ethyl benzoate towards the stationary phase.
Among column A and B, Column B give better separation of the components because both components have a similar boiling point.
