Simple staining requires one stain and a decolourizing agent.Its value lies in its simplicity and ease of use.After prepartion of the smear followed by heat fixing,it is covered with stain for a particular length of time.Excess of the stain is washed away with water and blots the slide dry. simple staining helps in observing the size,shape and arrangement of bacteria. Some stains that are used in simple staining procedure are crystal violet,methylene blue..etc.

In Kirby-Bauer method,bothe types ,that is ,gram negative and gram positive bacterias can be used as test organism,eg:Staphylococcus which is gram positive and Pseudomonas which is gram negative.The zone width is the function of initial concentration of the antibiotic ,its soluabilityand also its diffusion rate through agar.after the plates has been prepared, appropriate antibiotic disks are placed on it.then the plates are immediately kept for incubation at 35 degree celsius .the width of the zone is measured after 16 to 18 hours.
