The effect of prednisone is attributed to prednisolone. After administration, prednisone is converted to prednisolone. Anti-inflammatory effects are complex, but via binding to cellular glucocorticoid receptors, prednisolone acts to inhibit inflammatory cells and suppresses expression of inflammatory mediators. Prednisolone is approximately four times more potent than cortisol but only one seventh as potent as dexamethasone. Prednisone appears to be well absorbed and converted to active drug in dogs. However, in horses and cats, administration of prednisone results in low systemic levels of the active drug prednisolone, either because of poor absorption of prednisone or because of a deficiency in converting prednisone into prednisolone.
Prednisone is a commonly used catabolic steroid that binds to cytoplasmic receptors and inhibits DNA synthesis. Prednisone is converted to the active form, prednisolone, in the liver. Patients with hepatic dysfunction should be treated with prednisolone rather than prednisone.
