  1. You aim to detect human protein PTEN. Answer to following:

*highlight correct answer

  1. Primary antibody is anti-PTEN antibody. TRUE / FALSE
  2. Can use of secondary antibody be avoided? Briefly explain your opinion.
  3. Secondary antibody is anti-primary antibody. TRUE / FALSE

Solutions

Expert Solution

Ans a) True - The PTEN is a human protein. The primary antibody is the one which has the capacity to bind directly to a specific protein and majorly helps in purification and analysis process. The PTEN is basically a tumor suppressor protein and in majority of the cancer, this protein is mutated. When PTEN is the protein, the primary antibody is anti-PTEN as this directly and specifically binds to the PTEN protein. The anti-PTEN helps in detection and analysis of presence of PTEN as it has the tendency to bind to the antibody directly.

Ans 2) Yes, the secondary antibodies can be avoided in this condition because in this very highly specific type of primary antibody is required for the detection. This means that for detecting the PTEN proteins in the humans, it can be achieved by using anti-PTEN monoclonal antibodies which acts as primary antibodies. They are highly specific when it comes to recognition of the epitope present on the protein.

Ans 3) True

The secondary antibody is the isotype of the primary antibody and hence it can be raised against the primary antibody. The secondary antibody is not able to bind to the protein directly, but it interacts through the primary antibody. This means that the primary antibody binds to the PTEN protein and secondary antibody is raised which is highly specific to the primary antibody and hence it acts as anti-primary antibody.

In order to express a recombinant protein that will be used for antibody production, you are given the choice to use either a genomic library or a cDNA library. Both are good libraries that have been screened to identify genes and proteins. You decide to use the cDNA library. This is because: a. Introns have been removed from the mRNA and the exons spliced together to create an open reading frame in the transcript. b. The cDNA contains the exons...
