To check whether ampicillin resistance gene is acquired or not we have to perform assay in which below given media will be used

Nutrient agar meduium containing appropriate amount of ampicillin antibiotic with proper technique.

After preparing this media spread the bacterial culture on the plate and look for the growth on the next day after 24 hr incubation. If there is growth of bacteria which suggests that bacteria succesfully acquired ampicillin resistance gene and if no growth observed than it means no ampicillin gene acquired.

To calculate the transduction frequency perform replica plate technique by plating sample on the Nutrient agar plate and than replicate it on the Nutrient agar with ampicillin and look for numbers of colonies.

No of colonies on the Nutrient agar plate 100

No of colonies on the Nutrient agar plate with ampicillin 20

For example if you find total colonies 100 on nutrient agar plate and 20 colonies on the nutrient agar plate with ampicillin than it will be 20% transduction percentage.
