Manual methods pack Red blood cells by doing centrifugation. This information can be obtained by performing manual HCT direct measurement method, or by an automated hematology analyzer .
*It measures percentage of RBCs in sample of whole blood ,so it indicates oxygen-carrying capacity .

*A person with anemia usually have low RBC count hence he have lower PCV.
* When dehydrated,Blood volume decreases--So have a higher PCV than normal
