a.) Mutations in the C-terminus kinase domain of Receptor Tyrosine Kinases (RTK) that abolishes (completely stops) its activity. Please note that the kinase domain is important for autophosphorylation of RTK.
b.) Mutation in βARR (β-arrestin) that decreases its affinity for the phosphorylated cytoplasmic tail of the G-protein Coupled Receptor (GPCR).
c.) Mutation in estrogen receptor that prevents it’s binding to the HRE (hormone response elements) in the nucleus.
