Eventhough the resolution of image provided by a Transmission electron microscope is high,there are certain limitations that makes it not suitable for viewing the motility of a live Treponema.Those limiting factor's are:

  • Instead of a glass lens,an electron beam is used, which is penetrated from above the specimen by using a magnetic lens and projected onto a detector.Since this electrons have a limited penetrating ability, it is not possible to obtain a 3-D view of the specimen.
  • Another important condition for viewing a specimen is that both the beam and speciment should be in a vacuum and the specimen should be very thin and dehydrated.The treatment procedure for making the specimen dehydrated do not allow for viewing a live microbe.Hence, it is not possible to view the motility of a live Treponema.

Dark-Field microscopy is the ideal method used for visualizing a live Treponema,which is otherwise not possible through conventional light microscopy.It can create high-contrast and high-resolution images of specimens, without using any stains and thus eliminating the chances of live specimens getting killed or compromised by the stains.
