Process of conversion of UMP to dTMP takes place in the following steps:

1. Formation of UDP from UMP through the enzyme nucleoside monophosphate kinase by the conversion of ATP to ADP.

UMP+ATP --> UDP + ADP.

2. The conversion of UDP to dUDP by the enzyme ribonucleotide reductase class1. This reduces the carbon at C2 position of the ribose sugar hence converting it to deoxyribose sugar.

UDP+NADPH--->dUDP+NADP+

3. Conversion of the dUDP to dUTP which is catalysed by nucleoside diphosphate kinase coupled to the subsequent conversion of ATP to ADP.

dUDP+ATP--->dUTP + ADP

4. The conversion of dUTP to dUMP by the enzyme dUTP pyrophosphatase.

dUTP +H2O ---> dUMP + PPi

5. The conversion of dUMP to dTMP catalysed by the thymidylaste synthase with N5,N10- methylenetetrahydrofolate as the methyl donor.

dUMP + NA,N10 - methylenetetrahydrofolate ---> dTMP + dihydrofolate.

In this way UMP is converted to dTMP.
