Please discuss the role of the protein 4WDI
4WDI is the code in the protein data bank database for the protein called G9C8. This plays an important role in presenting the antigents to the T Cells. It has also a role in pathways involved in Type 1 Diabetes. The insulin reactivity by a CD8+ T-cell clone, which is known to induce Type 1 Diabetes, is characterized by weak TCR affinity to a highly unstable pMHC. The G9C8 TCR was able to bind more strongly to a peptide altered at the C terminus, thus demonstrating the potential of this T-cell clone to be triggered by a more immunogenic target. These studies also suggest that that the interaction between the TCR and pMHC is most likely to be focused toward the C terminus of the peptide, which explains that there is difference in sensitivity between the C-terminally altered peptide ligands. .The stability of the MHC complex is always crucial in presenting the antigensThe deconstruction or remodelling of this can result in increasing the effectiveness of TCR (T Cell Receptor Binding) and in helping to dilineate the role of the T Cell antigen presentation. By studies it was proved that modifications in the side chains in this 10 mer G9C8 peptide. Hence, this G9C8 binding induces flexible peptide presentation in which the MHC can effectively “open the back door” to accommodate an extra C-terminal peptide residues, thus helping in enhanced T Cell activation.
